They observe that TALENs were more efficient than ZFNs in generating correctly targeted iPSC colonies, but all corrected iPSC clones showed no signs of mutations at the top-ten predicted off-target sites of both nucleases. These data open the way to generate autologous, functionally corrected granulocytes with the TALEN system.
Here, the scientists showed that limited low-dose irradiation (LDI) using either γ-ray or x-ray exposure (0.4 Gy) significantly enhanced HR frequency, possibly through induction of DNA repair/recombination machinery including ataxia-telangiectasia mutated, histone H2A.X and RAD51 proteins. LDI could also increase HR efficiency by more than 30-fold when combined with the targeting tools zinc finger nucleases, transcription activator-like effector nucleases, and clustered regularly interspaced short palindromic repeats.
In this report, the authors used the CRISPR/Cas9 system to integrate a transgene in the AAVS1 locus. They observed that the knock-in of AAVS1 has significantly reduced MBS85 expression and increased contractile force. These results should be take in account in the design of gene therapy strategy which involve knock-in of AAVS1.
The authors describe a protocol for the targeted integration of a doxycycline-inducible transgene expression system in a safe harbor site in iPSCs. This gene targeting strategy uses zinc finger nucleases (ZFNs) to enhance homologous recombination at the AAVS. This protocol could be also compatible with the use of TALEN or CRISPR.
