Here, the authors describe an efficient Cas 9 version (Cas9) fused to a dihydrofolate reductase (DHFR)-derived destabilization domain (DD) and used to control gene expression with addition of Trimethoprim (TPM). They successfully used dCas9 to replace OCT4 in human cell and demonstrate that cell differentiation can be induced.
